For those eager to experience the Samurai perspective right away, you'll need to wait until nearly the end of the first act. The early game focuses solely on Naoe, with Yasuke making his playable debut during the Temple of the Horseman Quest. He then becomes playable again for the Fire and Lightning Quest. Once you complete this quest and Act I, you'll gain the ability to switch between the two protagonists more freely.
